Course contents:
1.The methods of chromatography, the role, the choice and optimisation of column properties in gas, liquid and supercritical fluid chromatography, modern instrumentation, conditions of analysis and their optimisation. Advanced signal processing and signal evaluation from column techniques. Hyphenation to spectrometry techniques, mainly mass spectrometry. Electromigration separation methods. Sample preparation with help of separation techniques. (allowance 2/0)
a.Column in gas chromatography. Instrumentation of gas chromatography, optimisation of analysis. The efficiency of separation. GC signal processing. Retention data and databases.
b.Column in liquid chromatography. Instrumentation of high performance liquid chromatography, optimisation of analysis, choice of separation systems, efficiency of separation. HPLC signal processing. Retention data and study of spearation mechanism.
c.Column, instrumentation and optimisation in supercritical fluid chromatography, choice of separation systems and modificators, optimisation of analysis.
d.Coupling of column chromatography techniques with mass spectrometry, charakteristics of mass transfer in interface, various ionisation techniques for GC-MS, HPLC-MS and SFC-MS. Evaluation of spectral signal and identification of compounds.
e.Multidimensional separation and methods of comprehensive twodimensional gas chromatography GCxGC, coupled liquid and gas chromatography HPLCxGC, twodimensional liquid chromatography HPLCxHPLC, methods of signal evaluation and chemometrics approaches.
f.Electromigration separation methods and coupling CE-MS. Column materials and choice of electrolyte system, optimisation of conditions. Capillary electromigration methods - CZE, MEKC, CEC, ITP. Coupling of electromigration methods with preconcentration and focusing.
