Learning outcomes of the course unit:
The aim of these lectures is to describe how cells exploit signalling components to assemble the specific signalling pathways, which they require to communicate which each other or to adapt to changes of external environment. The components and properties of major cell signalling pathways will be characterized. Attention will be focused on the role of signalling pathways in control of gene expression and cellular metabolism. The second half of lectures will deal with cell cycle signalling system and cell death
Course contents:
Signalling pathways in prokaryotes. Two-component system (TCS). Evolution and TCS in eukaryotes. Basic principles of cell signalling. Characterization of signalling components: signalling molecules, receptors, second messengers, effectors, signalling complexes. Integration and amplification of signals. Basic classification and characterization of membrane receptors. Intracellular/nuclear receptors. Principle of transfer information: posttranslation modification of proteins and conformation coupling. Protein phosphorylation/dephosphorylation (protein kinases and protein phosphatases - characterization and classification), oxidation, methylation, acetylation,sumoylation and ubiqutination. Structural domains and interaction of proteins. Major signalling pathways (SPs): SPs associated with second messengers(Ca2+, cAMP, cADPR, InsP3, DAG,Ptd Ins4,5P2 - signalling, NO/cGMP); redox signalling, MAPK signalling, NF-kB signalling, JAK/STAT signalling, TGF-b/SMAD signalling and Wnt signalling. Cell signalling and apoptosis. Cell cycle control.
