Title of the thesis:Structural analysis of avocado fruit polysaccharides (Persea americana, Mill.)
Summary:The analysis of polysaccharide structures is of general interest but also of industrial relevance as the structure and properties of polysaccharides are closely related. Polysaccharide analysis requires specialized techniques, which differ from those methods used for the characterization of small molecules. Methylation analysis is the most widely used and highly effective method for structure determination of polysaccharides. Despite its advantages we also comes into the contact with shortcomings such as: loss of information and the problem with analysis of ketosis. Because of this, it is necessary to use another method for determining the structure of polysaccharides which is called reductive cleavage. The structure of polysaccharides in avocado was determinated in this thesis. Both mentioned methods were used for this analysis. Higher comfort with determining the structure of polysaccharides was achieved by comparing their results. However, integral part of this analysis was the determination of molecular weight polysaccharides and their distribution by GPC (gel permeation chromatography).
