|Title of the thesis:||Production of Redox Enzymes and Their Application in Cofactor Regeneration|
|Summary:||This bachelor thesis is focused on enzymatic regeneration of cofactors nicotinamide adenine dinucleotide (NAD+) and nicotinamide adenine dinucleotide phosphate (NADP+) using the enzyme glucose dehydrogenase. Glucose dehydrogenase belongs to the class of oxidoreductases and catalyzes the conversion of D-glucose to D-glucono-1,5-lactone using NAD(P)+ as a cofactor, which is reduced during the reaction and can be used as a cofactor for a different enzyme. The experimental part of this thesis was focused on the production of glucose dehydrogenase by recombinant bacteria Escherichia coli, on the purification of the produced enzyme by affinity chromatography (FPLC) and on its characterization. The affinity towards NAD+ and NADP+ was compared and pH profiles, temperature profiles and kinetic parameters of the isolated glucose dehydrogenase were determined.|
