Title of the thesis:Production of Industrially Interesting Enzymes
Summary:This Master´s thesis was focused on the production of diamine oxidase using methylotrophic yeast Pichia pastoris. This enzyme belongs to amine oxidases and catalyzes the oxidation of primary amines to the corresponding aldehydes, ammonia and hydrogen peroxide. Diamine oxidase was found in cells of various organisms, including humans, where its main function is to degrade histamine, whose excess causes serious complications. Its application potential is in the therapy, pharmaceutical and food industry. Gene encoding diaminooxidase was successfully cloned into P. pastoris strains with different phenotype under the control of a strong, methanol inducible AOX1 promoter. The productivity of both strains was compared in fermentation production. Produced diamine oxidase was characterized by determining optimal conditions, kinetic parameters and monitoring the effect of various additions on enzyme activity. Long term stability was determined by optimal storage of diamine oxidase.
