Annotation: Mikroprostredie nádorov je veľmi plastické a jeho súčasti aktívne reagujú na zmeny zapojením signálnych dráh vedúcich k adaptácii buniek na zmenené podmienky. Opakované vystavenie buniek hypoxii vedie často k smrti buniek a k rozvoju nekrotických oblastí, v okolí ktorých sa selektujú bunky schopné adaptácie na nové podmienky. Cieľom práce bude proteomická analýza prítomnosti a zloženia interaktómu CA IX v rôznych bunkových líniach a v rôznych mikroenviromentálnych podmienkach – normoxia/hypoxia, acidóza, opracovanie chemoterapeutikami. Budeme analyzovať prítomnosť a interakcie medzi jednotlivými komponentami parciálneho interaktómu. Vybrané bunkové línie budú testované na schopnosť tvoriť 3D modely – sferoidy, v ktorých budeme sledovať zloženie metabolónu/parciálneho interaktómu a vzájomné interakcie zúčastnených proteínov v závislosti od gradientu pH, kyslíka a nutričných látok.
Annotation in English: The tumor microenvironment is very plastic and its components actively react to changes by the involvement of signaling pathways leading to the adaptation of cells to altered conditions. Repeated exposure of cells to hypoxia often results in cell death and in development of necrotic areas around which cells capable of adaptation to new conditions are being selected. The aim of the thesis is to perform proteomic analysis of the occurrence and composition of CA IX interactome in various cell lines and under different microenvironmental conditions - normoxia/hypoxia, acidosis, treatment with chemotherapeutics. We will analyze the presence of separate components and their interactions within the partial interactome. Selected cell lines will be tested for their ability to form 3D models - spheroids where we will study the composition of metabolon/partial interactome and mutual interactions of participating proteins in dependence on the gradient of pH, oxygen, and nutrients.
