Molecular mechanisms of the new drugs affecting oxidative stress ? the important factor of the number of illnesses

Project description:In previous VEGA projects (2/2054/22, 2/5010/5) the series of original antioxidants derived from pyridoindol stobadine was prepared. In preliminary studies it was, in accord with the theoretical assumptions found, that some of the newly prepared compounds have, in comparison with the original one, higher anti-oxidative power, lower acute toxicity and do not exhibit undesirable effects. It was found, that these compounds exhibit protect the nervous tissue against the damage caused by hypoxia or re-oxidation. It can be expected, that development of drugs the similar properties could be valuable contribution to the therapy of neural diseases with the participation of oxidative stress. Integral part of the project is the introduction of an integral exploitation of new electrophysiological, behavioral, biochemical, physicochemical, electrochemical and morphological approaches to the study of CNS. Besides the other things, the extend of the damage of the brain of gerbils during the brain incident will be studied using methods of nuclear magnetic resonance (MRI and MRS). Also electrochemical analysis of the mechanism of the action of the new indole antioxidants with neuro-protective effects (cyclic volt ?amperometry, ESR).
